Objective:
To evaluate the comparative epidemiology of hospital-onset bloodstream infection (HOBSI) and central line-associated bloodstream infection (CLABSI)
Design and Setting:Retrospective observational study of HOBSI and CLABSI across a three-hospital healthcare system from 01/01/2017 to 12/31/2021
Methods:HOBSIs were identified as any non-commensal positive blood culture event on or after hospital day 3. CLABSIs were identified based on National Healthcare Safety Network (NHSN) criteria. We performed a time-series analysis to assess comparative temporal trends among HOBSI and CLABSI incidence. Using univariable and multivariable regression analyses, we compared demographics, risk factors, and outcomes between non-CLABSI HOBSI and CLABSI, as HOBSI and CLABSI are not exclusive entities.
Results:HOBSI incidence increased over the study period (IRR 1.006 HOBSI/1,000 patient days; 95% CI 1.001–1.012; P = .03), while no change in CLABSI incidence was observed (IRR .997 CLABSIs/1,000 central line days, 95% CI .992–1.002, P = .22). Differing demographic, microbiologic, and risk factor profiles were observed between CLABSIs and non-CLABSI HOBSIs. Multivariable analysis found lower odds of mortality among patients with CLABSIs when adjusted for covariates that approximate severity of illness (OR .27; 95% CI .11–.64; P < .01).
Conclusions:HOBSI incidence increased over the study period without a concurrent increase in CLABSI in our study population. Furthermore, risk factor and outcome profiles varied between CLABSI and non-CLABSI HOBSI, which suggest that these metrics differ in important ways worth considering if HOBSI is adopted as a quality metric.
294 Identification of MCAK Inhibitors that Induce Aneuploidy in Triple Negative Breast Cancer Models

OBJECTIVES/GOALS: Microtubule poisons, like Taxol, are used to treat triple negative breast cancer (TNBC) and may induce lethal aneuploidy in cancer cells. Patients initially respond, but often develop drug resistance. New targeted drugs that cause aneuploidy may be a valuable approach to therapy. One potential target is the Kinesin 13 MCAK, which limits aneuploidy. METHODS/STUDY POPULATION: TCGA and GSE47561 databases were probed for MCAK expression, and data was stratified by subtype and survival statistics. Knockdown studies were performed to test whether MCAK knockdown sensitizes cells to taxanes for cell proliferation and for induction of aneuploidy. FRET and image-based screens were used to identify MCAK inhibitors from small molecule inhibitor libraries. Inhibitors were then tested for functional effects in multiple cell-based assays and for clonal growth in colony formation assays. RESULTS/ANTICIPATED RESULTS: MCAK expression is upregulated in TNBC and associated with reduced overall survival. Knockdown of MCAK caused a two-to-five-fold reduction of the IC50 for Taxol in cancer cell lines, with no change in normal cell lines. Taxol treatment or MCAK knockdown increased aneuploidy induction, with no additive effect between the two. Our small molecule screen identified three putative MCAK inhibitors, which induced aneuploidy in both taxane-sensitive and taxane-resistant cells. These inhibitors also reduced clonogenic growth, and the most potent inhibitor, C4, caused an approximate five-fold reduction in the IC50 for Taxol in cell proliferation assays. DISCUSSION/SIGNIFICANCE: MCAK can serve as a biomarker of breast cancer prognosis. MCAK knockdown or inhibition sensitizes cancer cells to Taxol without affecting normal cells, making it a potential target in combination therapy. MCAK inhibitors also reduce growth as single agents in taxane resistant lines, giving them potential use as therapies in resistant disease.

Effects of beet western yellows virus on growth and yield of oilseed rape (Brassica napus)

Field trials were undertaken in Suffolk in commercial crops of autumn-sown oilseed rape cv. Capricorn during 1993/94, cv. Apex in 1994/95. Plots were artificially infected with beet western yellows virus (BWYV) using viruliferous Myzus persicae, giving 73 to 94% infection. Control plots had natural infection ranging from 0 to 17·8%. Destructive plant samples were taken from each of the infected and control plots throughout the seasons for growth analyses, and final yields were measured on 44 m2 areas combine harvested from each plot. The seed yields of infected plots were 26 and 11% lower than control plots in 1994 and 1995 respectively (P<0·001).
Harvested seed yields were shown to be inversely proportional to the area of the plot that was inoculated with BWYV. Infection significantly lowered the oil content in 1995 from 47·9 to 46·8% (P<0·001), and increased glucosinolate levels from 16·12 to 18·37 μmol/g (P<0·01). BWYV caused a significant reduction in plant height and in numbers of primary branches in the 1993/94 trial and had an effect on the dry weight of the leaves, stalks, racemes and pods at some sample dates in both seasons. Virus-testing of infected plants showed that BWYV was present in the pod wall, the septum and seed coat; two of 78 embryo samples also contained virus. It was concluded that BWYV can cause significant yield losses in those years in which there is a high incidence of virus in the overwintered crops.

Upon the outbreak of World War I in August, 1914, the Russian Imperial Government rightly suspected that for reasons both of geography and of political climate, the Grand Duchy of Finland posed a great danger on its northern flank. This was hardly surprising. Since 1899, that Government had done all it could to destroy die effective autonomy which Finland had enjoyed within the Russian Empire during the nineteenth century.
“Russification” of Finland took place in two stages. General Nikolai A. Bobrikov, Governor-General from 1898 to 1904, reduced the Finnish Diet to a consultative assembly, suppressed newspapers, and introduced the Russian language into the Finnish Senate and civil service. In 1901, he dissolved the Finnish army, which had existed since 1877, and sought unsuccessfully to conscript Finns into the Russian army. To be sure, after Bobrikov's assassination and the Russian revolution of 1905, Tsar Nicholas II agreed to exempt Finns from Russian army service, and, in 1906, permitted the modernization of the Diet and the introduction of universal suffrage.
